Once on the M4 in Wales, you continue to junction 33, signposted Barry Island, Penarth and Cardiff West I think.  Turn off left at the top and continue for about 5 or 6 miles (roughly) until you see a left ramp for Cowbridge, called the A4232 I think (or something like that; its not too important).  I don't need to be too precise about these as you cannot miss them.  When you reach the lights at the top of the ramp, take the far right hand lane (marked, on the road, the A48) and follow the signs for Cowbridge.  (In the big picture, this is effectively a right turn at the first roundabout and straight over the second.  This absurd complex of roundabouts is known as Culverhouse Cross.  If you get at all lost anyone you ask will know where Culverhouse Cross is). 

You are now on the A48.  You stay on this road now for approx. 6 miles.  You go through a small village called St Nicholas (watch your speed here, they are tyrants on this stretch of road for giving out tickets) then past a golf club on your right, then through another village called Bonvilston then on again, past a pub at the side of the road on the right, then after about another 2-3 miles or so you will see a huge telecom mast on the right.  About 1/4 mile further on, down a dip, there is a left turn signposted St Hilary.  Take this turn and keep on this, now winding road through the village and out the other side, now signposted St Mary Church.  The road leads you a twisty ride through St Hilary and then you have about a mile of single lane traffic, twisting and turning until you eventually reach a T junction.  Turn left at the junction, follow the road round a slight right bend then up a small hill, a sharp left bend, another small climb and very soon you will see a village sign on the left for St Mary Church , with Llan Fair underneath.  Take this left turn, fairly steep hill up into the village then at the top you will see a church in front of you.
